Cellaca PLX applications

With the Cellaca PLX platform, you can:

  • Quickly perform cell purity checks and viability at the bench with Cellaca PLX immunophenotyping kits which combine CD3, CD4, and CD8 surface marker detection with cell viability in a single multiplex assay.
  • Stain transfected cell lines with viability dyes to determine the percentage of transfected or transduced cells that are live/dead.
  • Perform routine viability measurements as well as apoptosis functional assays to determine cell health.

Cellaca PLX optimized assay reagents and kits

The Cellaca PLX system provides flow-like data with simple-to-use, mix, incubate, wash, and read reagent kits that provide immunophenotyping plus cell counts and viability multiplexing assays using disposable, low fluorescence consumables. Quickly perform cell counts, concentration, purity checks, and viability at the bench with Cellaca PLX immunophenotyping kits which combine CD3, CD4, and CD8 surface marker detection with cell viability in a single multiplex assay.

Low auto-fluorescent consumables

Cellaca PLX consumable slides provide a safe and self-contained environment for the sample which then does not require a decontamination procedure of the instrument at the end of each run. The slide holder, which is included with the instrument, can accommodate 4 slides or 8 samples per experimental run.

Software that makes multiplexing easy

Stress-free Cellaca PLX software comes with preloaded, step-by-step templates and methodologies to streamline multiplex analysis, providing a benchtop solution for quick, reliable measurements of cell counts and concentrations, surface marker staining, viability, and apoptosis. And you get customizable results for presentations, too.

FAQs

  • How much volume of my sample do I need to perform a count?

    The minimum loading volume of stained sample in the counting plate is 50 µl. Typically, you will stain your cell sample 1:1 with AOPI dual-fluorescent viability dye, so you can perform a cell count with as little as 25 µl of cell sample. For cell-based assays such as immunophenotyping or GFP/RFP, instead of the counting plate, you will use low-fluorescent background slides that only require 15 µl of sample. If you have a low-volume sample, you can perform a cell count in the slides as well.

  • Can I save the plate for later if I only have a few samples to count?

    Yes, load the counting plate with as many or as few samples as you want to count. The Cellaca software will allow you to indicate which wells contain samples you wish to count and the rest of the plate can be saved for later use.

  • Are the counting plates compatible with multi-channel pipettes?

    Yes, the counting plates are compatible with multi-channel pipettes, making it fast and easy to stain and prep your samples. The Cellaca is also compatible with liquid handlers and automation platforms and has an optional automation license add-on, making it easy to scale up your throughput.

  • What daily start-up or shut-down cycles do I need to run?

    There are no daily start-up or shut-down cycles for the Cellaca because there are no internal fluidics and nothing that needs to get cleaned or could clog. The Cellaca uses LEDs as a light source. The only prep you need to do is turn on the instrument and start up the software, and the Cellaca is ready to count your samples right away.

  • How does the Cellaca count challenging or messy cell types?

    The Cellaca is designed to identify live and dead nucleated cells, even in very messy samples with RBCs and debris or in irregular-shaped cell populations. By using the AOPI dual-fluorescence viability dye, only nucleated cells are counted and RBCs and debris are ignored (this method is so specific, it even works in whole blood without lysing!). The Cellaca will show you the images it captured and which cells were counted so that you can visually verify the results and trust your data.
