The study of antibody internalization is important in the research and development of antibody-drug conjugates (ADCs), a rapidly expanding class of biopharmaceuticals that link a monoclonal antibody to a cytotoxic payload via a cleavable linker. Once an antibody or ADC binds its target on the cell surface, the complex is endocytosed and routed to acidic endo-lysosomal compartments where the payload is released, triggering cell death through mechanisms such as DNA damage or microtubule disruption. Effective methods to quantify this internalization in live cells are important in ADC development and for understanding receptor biology.
