For receptor internalization experiments
The signals measured at 620 nm are used to plot the sigmoidal agonist dose-response curves, from which half maximal effective concentration (EC50) values are calculated (potency of the agonist response).
A "fold-of-change" (FOC) ratio is calculated by dividing the signal at 620 nm obtained at each agonist concentration by the signal at 620 nm obtained in the absence of agonist (medium-only condition). The FOC is used to evaluate the amplitude (efficacy) of the response.
For more details, please refer to the manuals of the pHSense™ reagents.
For antibody/ADC internalization experiments
Specific signals are calculated by subtracting the negative (irrelevant) control antibody signal at 620 nm from the test antibody signals at 620 nm. The specific signal is used to plot the sigmoidal agonist dose-response curves, from which half maximal effective concentration (EC50) values are calculated (potency of the agonist response).
A "fold-of-change" (FOC) ratio is calculated, by dividing the signal at 620 nm obtained with the test antibody by the signal at 620 nm obtained with the pHSense Fab only control. The FOC is used to compare various antibodies and various assay test conditions.
