An innovative approach for direct detection of intracellular proteins in human whole blood using AlphaLISA SureFire® Ultra technology.

This application note describes a no-wash approach for detecting total and phosphorylated intracellular proteins in human whole blood using AlphaLISA™ SureFire® Ultra™ immunoassays. The workflow overcomes traditional matrix and handling challenges, enabling sensitive biomarker measurement with minimal sample processing.

In this application note, you will access:

  • A simplified workflow for measuring total and phosphorylated intracellular proteins directly in whole blood
  • Data demonstrating robust analysis of MAPK and JAK/STAT signaling pathways, including PROTAC mediated protein degradatio
  • Practical guidance on selecting the appropriate model (whole blood, PBMCs, or primary macrophages) for low abundance targets such as IRF5
