HMGB1, IL-1β and IL-18 assays used together to discriminate pyroptosis and necroptosis

Whereas apoptosis is a low inflammatory cell death, necroptosis and pyroptosis are highly inflammatory due to cell membrane permeabilization and cause the release of Danger Associated Molecular Patterns (DAMPs). Among the DAMPS is HMGB1, which induces an immune response. HMGB1 has been associated with several diseases like atherosclerosis, arthritis, or sepsis.

To help you discriminate between necroptosis and pyroptosis, Revvity has prepared this note with:

  • HTRF HMGB1 assay principle and its protocol,
  • How, when associated with our IL18 and IL6 kits, it enables discrimination between necroptosis and pyroptosis,
  • Comparison with luciferase and ELISA assays.
