Can better control of transgene expression improve rAAV production?
rAAV production can be challenging when transgenes cause stress or toxicity in virus-production cells. Even low-level premature expression can reduce cell viability and limit vector output.
Revvity scientists combined the HEKcellence™ platform, a HEK293 virus-production cell system, with Vectonize™ technology, an shRNA-based platform that suppresses transgene expression during vector production. HEKcellence provided higher full capsid yields across multiple rAAV serotypes, while Vectonize reduced premature transgene expression during packaging.
The combined approach increased vector output across several challenging payloads, including up to a 76-fold increase in titers for a toxic transgene with AAV2. Higher virus-production cell viability was observed without reducing transgene potency following transduction.
