Beyond broad tropism: targeted lentiviral vectors through surface modification

Can lentiviral vectors be targeted to specific cell types while maintaining manufacturability?

Standard VSVG-pseudotyped lentiviral vectors have broad cellular tropism, which can result in non-specific targeting. Some therapeutically relevant cells, including natural killer cells, also lack VSVG receptors and remain inaccessible to standard LV vectors. Alternative glycoproteins can refine tropism but may affect particle stability and manufacturing.

Revvity’s surface modification platform combines VSVG with a cell-specific binding domain, such as an ScFv or nanobody. Parameters including VSVG ratio, linker length and sequence can be adjusted while using existing manufacturing workflows.

In a proof-of-concept study targeting CD3-expressing cells, modified LV particles produced in both adherent and suspension systems targeted the T-cell model more efficiently than the control cell model. Particle manufacturability remained consistent across the linker variants tested, and nanobodies were also compatible with the platform.
